Our lab has a broad spectrum in questions concerning reproductive biology and immunology. We investigate Ion- and proton channels as crucial molecules enabling sperm to regulate all of their key functions like motility, hyperactivation or sperm-egg communication. These questions are addressed with the help of photometry in combination with pharmacological protocols. We also use all classical and modern methods of cell imaging (Immunocytochemistry electron microscopy, digital holographic microscopy), molecular biology and protein biochemistry in our lab. Our recent work concentrates on the spatial-temporal imaging of sperm movement with the help of holographic microscopy.
Most would agree that life starts with the fusion of egg and sperm. Millions of sperm are rereleased into the oviduct after ejaculation. Few have considered how the fertilizing sperm is selected from the millions of sperm released to the female genital tract. During their way along the oviduct – a journey that is equivalent to a human walking from Frankfurt to Berlin within 24 hours – many selective processes decide which sperm are possible candidates to fertilize the egg. No more than 10-20 sperm will come even close enough to have a chance to fertilize the oocyte. We plan to apply the cutting-edge technique of digital-holographic Microscopy (DHM) on sperm movement to analyze their trajectories and flagellar wave in four dimensions. We have developed this technique in the last years that far, that we cannot apply only a head tracking of the sperm but observe the whole flagellar movement in space and time in all dimensions. This with a spatiotemporal resolution which is only limited by the speed of the camera used. We could show that murine sperm have a well-defined chiral movement in the head as well as in the flagellum, which work independently from each other. In our ongoing projects we plan to apply DHM to certain and well-defined points during fertilization. Those time points will be early activation, linear movement, capacitation, sperm-egg binding/fusion, and hypermotility. We choose these time points, because they are very well defined and can easily be simulated in-vitro. We will perform these experiments in part with the help of transgenic mice which carry mutations for the CatSper channel. CatSper is across many species one of the most important ion channels for successful fertilization. We will also use sperm of other species beside mouse – like bovine, human and sea urchin to make conclusion about translational importance of our findings.We will further identify the importance of the above mentioned chiralities of the sperm head and flagellum in the light of different key functions of spermatozoa. We will specifically test the hypothesis, if altering chiralities of the head and flagellum are important for the regulation of sperm functions. We expect to generate significant new findings for the basic biological knowledge of fertilization as well as information about important factors which can lead to disturbance of sperm movement and in turn to difficulties of sperm selection during fertilization.
Das Leben beginnt mit der Fusion von Spermien und Eizelle. Millionen von Spermien werden durch die Ejakulation in den Ovidukt entlassen. Die Spermien legen danach innerhalb von 24h einen Weg zur Eizelle zurück, der einer Wanderung von Frankfurt nach Berlin entspricht. Selektionsprozesse entscheiden auf diesem Weg, welche Spermien soweit in die Nähe der Eizelle gelangen, dass diese überhaupt eine Chance haben, sie zu befruchten. Nicht mehr als 10-20 Spermien bleiben am Ende dieses Selektionsprozesses übrig. Wir planen mit Hilfe der digital-holographischen Mikroskopie (DHM) Trajektorien und Wellenbewegungen von Spermien bzw. deren Flagellen zu untersuchen. Die Methode der DHM wurde von uns so weit entwickelt, dass wir als erste Arbeitsgruppe nicht nur ein Kopf-Tracking, sondern die Bewegung des gesamten Spermiums mit Flagellum in Raum und Zeit verfolgen können. Dies mit einer spatiotemporalen Auflösung, die lediglich durch die Kamerageschwindigkeit limitiert wird. Dabei konnten wir zeigen, dass Spermien sowohl im Kopf als auch im Schwanz fest definierte Chiralitäten zeigen, die unabhängig voneinander gesteuert werden. In unseren derzeitigen Untersuchungen sollen zu definierten Zeitpunkten der Fertilisierung Schlüsselfunktionen von Spermien mittels DHM untersucht werden. Wir wählen dazu die Zeitpunkte der der frühen Aktivierung, der linearen Motilität, der Kapazitation, der Spermien-Eizellbindung und der Hypermotilität, da diese sich in vitro klar definieren bzw. simulieren lassen. Wir werden dies u.a. mit Hilfe von transgenen Tieren für CatSper durchführen, da dieser Ionenkanal speziesübergreifend von großer Bedeutung für die Befruchtung ist. In weiteren Untersuchungen werden humane, murine, bovine und auch Spermien von Seeigeln zum Einsatz kommen, um die translationale Bedeutung der von uns gemachten Beobachtungen zu beurteilen. Mit unseren Experimenten soll die Bedeutung der oben erwähnten Chiralitäten von Kopf und Flagellum unter dem Einfluss unterschiedlicher Spermienfunktionen untersucht werden. Wir möchten insbesondere die Hypothese testen, ob sich ändernde Chiralitäten von Kopf und Flagellum wichtig für Steuerung von Spermienfunktionen sind. Wir erwarten neben dem Erkenntnisgewinn zum biologischen Grundverständnis der Spermienbewegung Informationen darüber, welche Faktoren zu Störungen der Spermienbewegung führen können und so über die Selektion von Spermien während der Befruchtung entscheiden.

Male reproduction
All areas of signaling in reproduction are of interest to our group.
We identify and characterize molecules important for spermatogenesis, early and late activation of spermatozoa and cell-egg interaction. We also focus on cell-cell interaction in testis and prostate cells.
Female reproduction
This project focuses on the detailed cellular mechanisms in female reproduction and their function and dysfunction in mouse and human. This includes therapeutical approaches to endometriosis, gene regulation of feto-maternal interaction and oocyte development and oocyte aging.
Prostate cancer
microRNAs play an important role in post transcriptional regulation of the gene regulatory pathways of most cells. This project focuses on the importance of microRNAs for prostate cancer development and progression in animal models as well as in in-vitro experiments.
CEACAMs
This work deals with the role of CEACAMs in infection, inflammation and wound healing. We characterize the CEACAM mediated cell-cell and cell-pathogene communication, signaling and function. Monoclonal antibodies, glycoproteins, celllines and mouse are generated as tools.

Die Infertilität von Paaren ist ein weltweites Problem, dass in 40-50% der Fälle auf Fertilitätsstörungen beim Mann zurückzuführen ist. Eine grundlegende Ursache für die männliche Infertilität ist ein Defekt in der Spermienmotilität. Obwohl einige, für die Spermienmotilität wichtige Signalwege bekannt sind, ist die vierdimensionale Spermienschwimmbewegung und deren Bedeutung für die Fertilisation der Oozyte noch nicht vollständig verstanden.
Der Forschungsschwerpunkt von Frau Dr. Wiesehöfer liegt daher in der vierdimensionalen Bewegungsanalyse von Spermien mittels der digital-holografischen Mikroskopie (DHM). Mithilfe dieser Technik sind wir erstmals in der Lage neben der Schwimmtrajektorie auch die Bewegung des Spermienschwanzes (Flagellums) in 4D darzustellen. Die Verwendung von spezifischen Messkammern erlaubt uns Bewegungsanalysen unter definierten Versuchsbedingungen durchzuführen. Der Einfluss verschiedener Faktoren auf die 4D Spermienbewegung kann mittels einer Perfusionskammer untersucht werden. Des Weiteren ermöglicht ein in das DHM inkludiertes Fluoreszenzmodul die gleichzeitige Analyse biochemischer Veränderungen, die während des Fertilisationsprozesses auftreten. Um neue Erkenntnisse zum biologischen Grundverständnis der Spermienbewegung, insbesondere der Flagellenbewegung kurz vor der Befruchtung, zu erhalten, untersucht Frau Dr. Wiesehöfer die Spermienmotilität zu definierten Fertilisationszuständen (aktiviert vs. hyperaktiviert) sowie nach Kontakt zur Zona Pellucida, einer Glykoproteinmatrix, die die Eizelle umgibt. Zusätzlich wird der Einfluss verschiedener Ionenkanäle (CatSper, Hv1), sowie Änderungen des intrazellulären pH-Werts auf die vierdimensionale Spermienbewegung untersucht. In diesem Zusammenhang werden weitere Methoden wie High-speed Imaging und Computer-Assistierte Spermienanalyse (CASA) verwendet.
Infertility of couples is a worldwide problem, which depends in 40-50% of these cases on the fertility dysfunction of men. Although some causes of male infertility and some important signaling pathways of sperm motility are already known, the four dimensional sperm movement and its meaning for the fertilization process of the oocyte is not yet fully understood.
The research focus of Dr. Wiesehöfer relies on the four dimensional movement analysis of sperm using digital-holographic Microscopy (DHM). Besides the analysis of sperm trajectories, this technique allows the analysis of the sperm tail (flagellum) movement in 4D for the first time. The use of specific measurement chambers enable us to perform motility analysis under defined trial conditions. The influence of different factors on the 4D sperm movement can be analyzed with the use of perfusion chambers. Furthermore, a fluorescence module included to the DHM allows us a parallel analysis of biochemical changes, which occur during the fertilization process. To gain new insights into the biological understanding of sperm motility especially of flagellar movement, Dr Wiesehöfer examine the sperm motility to defined fertility conditions (activated vs. hyperactivated) and after contact to the zona pellucida, a glycoprotein matrix, which surrounds the oocyte. Additionally, the influence of different ion channels (CatSper, Hv1) as well as changes of intracellular pH on four dimensional sperm movement will be analyzed. In this context, other methods like high-speed imaging and computer-assisted sperm analysis (CASA) will be used.

Der Forschungsschwerpunkt von Dr. Irina Kube-Golovin umfasst die Tumorbiologie und Immunologie mit besonderem Fokus auf die Signaltransduktionsmechanismen von CEACAMs (Carcinoembryonic Antigen-related Cell Adhesion Molecules). Diese Familie von Zelladhäsionsmolekülen spielt eine zentrale Rolle in biologischen Prozessen wie Zellkommunikation, Immunmodulation und Tumorentwicklung. CEACAMs befinden sich vor allem auf der Oberfläche von Leukozyten sowie auf Epithel- und Endothelzellen. Durch ihre Interaktionen regulieren sie die Zellkommunikation und die Signaltransduktion in verschiedenen biologischen Kontexten. Dr. Kube-Golovin fokussiert sich insbesondere auf die Rolle von CEACAMs in der Tumorentstehung und -progression. Spezifische CEACAMs sind dabei als potenzielle Biomarker und therapeutische Targets von Interesse, da sie aktiv an der Tumorbildung und -ausbreitung beteiligt ist. Ziel der Forschung ist es, die CEACAM-vermittelten Signalwege besser zu verstehen und deren Einfluss auf Tumorentwicklung sowie Immunantwort zu entschlüsseln, um neue, gezielte therapeutische Ansätze zu entwickeln.
Zusätzlich widmet sich Dr. Kube-Golovin der Forschung zu extrazellulären Vesikeln (EVs) und den therapeutischen Einsatzmöglichkeiten von Nanomaterialien. Extrazelluläre Vesikel sind entscheidend für den interzellulären Informationsaustausch und die Modulation von Immunantworten, weshalb sie eine vielversprechende Zielstruktur in der Tumorforschung darstellen. Zudem untersucht Dr. Kube-Golovin den Einsatz von 2D Nanomaterialien (MXenes), um gezielt auf CEACAMs und andere Tumormarker einzuwirken und Tumorzellen selektiv zu zerstören.
Die interdisziplinäre Forschung vereint Ansätze aus Zellbiologie, Molekularbiologie, Immunologie und Materialwissenschaften und trägt dazu bei, neue Strategien zur Bekämpfung von Tumorerkrankungen zu entwickeln. Ein zentraler Bestandteil sind eigenständig entwickelte molekulare Werkzeuge, wie zum Beispiel spezifische Antikörper und rekombinante Proteine, die es ermöglichen, präzise Wechselwirkungen auf molekularer Ebene zu untersuchen. Darüber hinaus kommen verschiedene Zell- und Mausmodelle zum Einsatz, die als wichtige Plattformen dienen, um die in-vitro gewonnenen Ergebnisse auf die Komplexität eines ganzen Organismus zu übertragen und zu validieren.
Dr. Kube-Golovin betreut und bietet Bachelor-, Master- sowie Doktorarbeiten für naturwissenschaftliche und medizinische Studierende an. Die Beteiligung an EU- / DFG-geförderten Projekten und Kooperationen mit nationalen sowie internationalen Partnern bieten interdisziplinäre und abwechslungsreiche Möglichkeiten für wissenschaftliche Zusammenarbeit, den Austausch von Wissen und die Weiterentwicklung innovativer Forschungsansätze.
Research Focus: Tumor Biology, Immunology, Signal Transduction, Extracellular Vesicles, Nanomaterials
Dr. Irina Kube-Golovin’s research focus includes tumor biology and immunology, with a particular emphasis on the signal transduction mechanisms of CEACAMs (Carcinoembryonic Antigen-related Cell Adhesion Molecules). This family of cell adhesion molecules plays a key role in biological processes such as cell communication, immune response, and tumor development. CEACAMs are primarily expressed on the surface of leukocytes as well as on epithelial and endothelial cells. Through their interactions, they regulate cell communication and signal transduction in various biological contexts. Dr. Kube-Golovin focuses on the role of CEACAMs in tumor formation and progression. Specific CEACAMs are of particular interest as potential biomarkers and therapeutic targets, as they are actively involved in tumor development, progression and spread. The research aims to gain a deeper understanding of CEACAM-mediated signaling pathways and their effects on tumor development and immune response, with the goal of developing new, targeted therapeutic strategies.
In addition, Dr. Kube-Golovin focuses on the research of extracellular vesicles (EVs) and the therapeutic potential of nanomaterials. EVs are crucial for intercellular communication and modulation of immune responses, making them a promising target in cancer research. Furthermore, Dr. Kube-Golovin investigates the use of 2D nanomaterials (MXenes) to selectively target CEACAMs and other tumor markers, aiming to selectively destroy tumor cells.
Interdisciplinary research combines approaches from cell biology, molecular biology, immunology, and materials science to develop new strategies for combating cancer. A key component of this research is the use of self-developed molecular tools, such as specific antibodies and recombinant proteins, which enable precise investigation of molecular interactions. In addition, various cell and mouse models are employed as important platforms to transfer and validate in vitro findings within the complexity of a whole organism.
Dr. Kube-Golovin supervises and offers Bachelor’s, Master’s, and doctoral theses for students in the natural sciences and medicine. Participation in EU- / DFG-funded projects and collaborations with national and international partners provides interdisciplinary and diverse opportunities for scientific cooperation, knowledge exchange, and the further development of innovative research approaches.

Das Prostatakarzinom ist weltweit immer noch eine der häufigsten diagnostizierten Tumorarten bei Männern und ist aufgrund der hohen klinischen Relevanz ein Schwerpunkt der Tumorforschung. Die Ursachen für die Entstehung dieser Tumorentität sind noch nicht vollständig geklärt, jedoch konnte die Fehlexpression bekannter Onkogene und Tumorsuppressoren sowie nicht-kodierender RNAs beobachtet werden. Der Forschungsschwerpunkt von Dr. Dankert liegt auf der Untersuchung grundlegender Expressionsänderungen proteinkodierender Gene und verschiedener nicht-kodierender RNA-Spezies im Prostatakarzinom sowie ihrer Charakterisierung. Hierbei möchte er neue Gene sowie RNAs identifizieren, die einen Einfluss auf die Entstehung und Progression dieser Tumorentität haben, um weitere Ansatzpunkte für zukünftige Therapien zu finden sowie mögliche Diagnostikverbesserungen zu erhalten.
Im Labor kommen eine Vielzahl von Methoden aus der Molekularbiologie, Proteinbiochemie und Zellbiologie zum Zuge. Dazu gehören Klonierungen von Genen, Reportergenassays, Expressionsanalysen mittels qRT-PCR, Micro Arrays oder Deep Sequencing, Zellkultur-basierte funktionelle Untersuchungen von Genkandidaten. Neben dieser in vitro Methoden werden zusätzlich in situ Analysen wie z.B. Immunfluoreszenz- sowie immunhistologische Färbungen von Gewebe sowie RNA-ISH durchgeführt.
Prostate carcinoma is still one of the most commonly diagnosed cancers in men worldwide and stays in the focus of cancer research due to its high clinical relevance. The genesis for this tumor entity hasn’t been fully clarified yet, though a deregulated expression of oncogenes and tumor suppressors as well as non-coding RNAs are described. The focus of Dr. Dankert lies on the analysis of expression changes concerning protein-coding genes as well as non-coding RNA species and their functions in prostate carcinoma. Here, he wants to identify new genes and RNAs, which have an impact on tumorigenesis and progression, aiming at new therapy starting points and improvement of diagnostics.
For this, a multitude of methods from molecular biology, protein biochemistry and cell biology is used. They include gene cloning, reporter gene assays, expression analysis by qRT-PCR, micro arrays and deep sequencing as well as cell-based functional analyses of candidate genes. Beside in vitro methods, Dr. Dankert also performs in situ analysis such as IF and IHC stainings or RNA-ISH.

Das Prostatakarzinom ist aktuell die zweithäufigste Tumorerkrankung bei Männern weltweit und aufgrund der hohen Anzahl an Sterbefällen ein Schwerpunkt der Tumorforschung. Patienten mit einem fortgeschrittenem Prostatakarzinom werden mit einer Androgen deprivierenden Therapie behandelt, die allerdings nach Langzeitbehandlung oft zu einem nicht mehr behandelbaren Tumorrezidiv führt. Ein Grund dafür, ist die neuroendokrine Differenzierung von Prostatakarzinomzellen, welche Androgen-unabhängig die Tumorprogression fördert. Der Forschungsschwerpunkt von Dr. Wiesehöfer beruht auf der Untersuchung dieses Prozesses sowie der Eigenschaften der Neuroendokrin-ähnlichen Zellen. Hierbei möchte er Gene und nicht-kodierende RNAs identifizieren, die einen potenziellen Einfluss auf die neuroendokrine Differenzierung haben, um mögliche Therapieansätze und neue Diagnostikverfahren zu entwickeln.Eine relativ neue Therapieform ist die Immuntherapie, welche der immunsupprimierenden Wirkung der Tumorzellen und der damit verbunden Immunevasion, entgegenwirkt. In Zukunft soll das Tumormikromilieu und die Mechanismen der Immunevasion von Prostatakarzinomzellen untersucht werden.Für dieses Ziel bedient sich Dr. Wiesehöfer einer Vielzahl von Methoden aus der Molekularbiologie, Proteinbiochemie und Zellbiologie. Zu den molekularbiologischen Methoden zählen Gen-Klonierungen und verschiedene Methoden der Expressionsanalyse. In der Proteinbiochemie werden immunologische Färbungen von Geweben und Zellen angefertigt. Funktionelle Untersuchungen von Genen/ Proteinen werden mit Hilfe verschiedener in vitro Analysen, wie z.B. der Durchflusszytometrie durchgeführt.
Prostate carcinoma is currently the second most common tumor disease in men worldwide and, due to the high number of deaths, a focus of tumor research. Patients with advanced prostate carcinoma are treated with androgen-depriviating therapy, which, however, often leads to an untreatable tumor recurrence after long-term treatment. One reason for this is the neuroendocrine differentiation of prostate cancer cells, which promotes androgen-independent tumor progression. The research focus of Dr. Wiesehöfer is based on the investigation of this process as well as the properties of neuroendocrine-like cells. Here, he wants to identify genes and non-coding RNAs that plays a potential role in neuroendocrine differentiation in order to develop possible therapeutic approaches and new diagnostic tools.Immunotherapy is a relative new treatment method, which counteracts the immunosuppressive effect of tumor cells and the associated immune evasion. In the future, the tumor microenvironment and the mechanisms of immune evasion of prostate carcinoma cells will be investigated.For this aim, Dr. Wiesehöfer uses a variety of molecular biology, protein biochemistry and cell biology methods. Molecular biological methods include gene cloning and various methods of expression analysis. In protein biochemistry, immunological staining of tissues and cells is prepared. Functional investigations of genes/proteins are performed with different in vitro analyses, e.g. flow cytometry.
